Plant regeneration of fenugreek (Trigonella foenum-graecum L.) plant from cotylednory nodes callus

AbstractIn this study, callus cultures of fenugreek (Trigonella foenum-graecum L.) was produced easily from hypocotyl, of sterile two weeks old seedlings, these explants were divided into three pieces, the piece near the top of the cotyledon node given the symbol C1, the others near the crown area C2 and C3 represented the cotyledon node. These pieces cultured in MS (Murashige and Skoog) medium supplemented with of 2.0 mg. L-1 BA + 1.0 mg L-1 NAA or 0.4 mg L-1 TDZ + 0.1 mg L-1 IBA or 0.5 mg L-1 NAA + 15% Coconut Water. The results showed the ability of cotyledon node callus to produce plantlets in one step regeneration in the induction medium after 15 day from reculture. Callus produced from nodes proliferated numerous shoot on agar solidified MS medium supplemented with 0.5 mg. L-1 NAA + 15% Coconut Water. The results of this study showed no shoot regeneration from C1 and C2 callus produced from hypocotyl.
