Abstract
The present study is conducted to in restigate the bacteria Pseudomonas aeruginosa available in 50 samples of soil taken from different location in the city. 50 samples of water are combined. It is also investigated for the presence of bacteria in 50 patient of urin samples and wounds collected from hospitalized patients and visitors , to the Ramadi general hospital , possible to examine 94 isolated as a subsidiary of bacteria. This study also includes a determination of the isolates' ability to form the biofilm and estimate the amount of protein in the extract of biofilms formed. The effect of using the volume of oculum bacterial is studied in the growth of these bacteria and biofilm production . The study intrudes a determination the ability of isolates growth and the formation of biofilms in the different pH( natural , acidic ,alkaline). The effect of temperature is also studied in the growth of the isolates and the formation of memberance with different range temperature of ( 5 – 65 ) C. The study indicates the effect of the processes of stirring and aeration on the growth and biofilms production in four session ( 50, 100 , 150 ,200 ) rpm. Then ,the efficiency of isolates are tested in representing the carbonic and nitrogen resources used in different concentration(1-5)% in the growth isolates and biofilms production .
The most important results are : Bacteria P. aeruginosa are isolated 94 isolates are distributed according to the samples taken from sites – 33 isolates are obtained from the total number of the samples . the percentage of water samples acquired is 29 bacterial isolates have been obtained .While the percentage of the bacteria isolated from the total patient samples is 32. The efficiency of bacterial isolates is varied in their formation to the biofilm is ranging between (1.1 – 6.5 )mm. five local isolates of bacteria P . aeruginosa are selected and they occupy the numerals( 6 , 49 , 81 , 69 , 94 ) this is by considering them the best and the most efficient in the formation of biofilm . The efficiency nosa is varied and they carry the numerals ( 6 , 49 , 81 , 69 , 94. The highest concentration is recorded in the biofilm of the local isolates P . aeruginosa 81 which reaches 0.12 mg.ml-1 . in contrast the lowest concentration of protein in the biofilm of the local isolate is P . aeruginosa 6 which reaches 0.035 mg.ml-1. It is indicated that the best volume of added bacterial vaccine is 1 ml of leads to the increase in the growth and the formation of biofilm. the best growth of isolatesis obtained at pH 7.2 ,while the best pH for the formation of biofilm of the these isolates is( 6.2 , 9.2 and 10.2 ) depending or the thickness of biofilm. The best temperature to the growth of these isolates is 35 c◦ . On the other hand , the best temperature to form the biofilm is 45c◦. The process of stirring and airing of the media leads to the increase of growth and the formation of biofilms to reach at 11.633 Log cfu.ml-1 at 150 rpm and thickness of membrane 3.333 mm. at the same speed of stirring. The best growth to these isolates with the carbonic-nitrogen source in which lactose with( NH4)2SO4,while are found that the local isolate P.aeruginosa 81 grows well under the effect of the nitrogen and carbonic sources and also the formation of biofilm in a good way under the effect of the sources mentioned above.
