Evaluation of Immunostrip-ELISA Test for Identification of TMV and CMV Viruses in IRAQ

AbstractThis study was conducted to evaluate the efficacy of Flashkits to apply Immunostrip ELISA test to identify Cucumber mosaic virus (CMV) and Tobacco mosaic virus (TMV) in Iraq. Samples from Cucumber, Melon, Winter squash, and Zucchini showed symptoms suspected to be CMV, also samples from Tomato with strip leaves symptoms and Tobacco with mosaic symptoms suspected to be TMV, were collected from different fields of Baghdad area. Results of bioassay on the indicator plants, inoculated by extracts from leaves of infected Cucumber, Melon, Winter squash and Zucchini plants on Cucumber, Cowpea, Chenopodium, Tomato, Melon, Winter squash, Zucchini, Datura, Tobacco and Water melon indicates the presence of CMV. Results of bioassay on the indicator plants, Cucumber, Cowpea, Tomato and Tobacco (Xanthi & Turkish) inoculated by extracts from leaves of infected Tomato & Tobacco plants indicates the presence of TMV. Results of Immunostrip ELISA specific for CMV, in extracts from Cucumber, Melon, Winter squash and Zucchini plants showed a line of precipitation which indicates infection by CMV. Also, a line of precipitation on the Flashkits specific for TMV was appeared, in extracts from tomato and tobacco which indicates the presence of TMV. These results were in accordance of bioassay results indicates that the Immunostrip-ELISA was efficase with high resolution for identifying of Iraqi CMV and TMV isolates.
